SPONTANEOUS cases of Creutzfeldt-Jakob disease may be triggered when a protein ends up in the wrong part of brain cells. In CJD, a protein called PrP is folded wrongly and forms insoluble clumps. Now Jiyan Ma and Susan Lindquist of the University of Chicago in Illinois have found that when yeast cells are genetically engineered to produce normal PrP in their endoplasmic reticulum—the space in cells where the protein usually folds—some PrP gets misdirected into the cytoplasm and forms clumps like those seen in the brains of CJD patients (Nature Cell Biology, vol 1, p 358).
